Detailed introduction to the use of CO2 incubators
The CO2 incubator is improved on the basis of ordinary cultivation, mainly by adding CO2 to meet the environment required for cultivating microorganisms. It is mainly used for tissue culture and the cultivation of some special microorganisms.
1. Installation and debugging of CO2 incubator
There is an operation panel on the front of the CO2 incubator, on the panel there are power switch, temperature regulator (manual and LCD panel), injection switch, adjustment knob, temperature display panel, display panel and humidity display panel, sampling hole (for Take a sample and check whether the CO2 in the incubator reaches the content displayed on the display panel) and the alarm device (over-temperature alarm light).

(1) The CO2 incubator should be placed in a stable location away from heat sources to prevent temperature fluctuations and microbial contamination.
(2) Before turning on the power, add a certain amount of distilled water into the incubator according to the instruction manual (add a certain amount of disinfectant, see the instruction manual for details) to avoid burning out the machine.
(3) When water is added to a certain amount, the alarm light is on, that is, it stops, turn on the power switch, start heating, and adjust the thermostat to the required temperature.
(4) When the temperature reaches the desired temperature, the heating will stop automatically. When the desired temperature is exceeded, the over-temperature warning light will illuminate and an alarm will sound.
(5) The CO2 used in the CO2 incubator can be liquid or gas. No matter which one is used, the pipeline should not be bent too much to ensure the smooth flow of gas. Typically, gas cylinders are selected and connected to pressure control gauges.
(6) Return the carbon dioxide content to zero. When the temperature and humidity in the cabinet are stable (usually three days), turn the adjustment knob to adjust the number of display panels to 0.00, and repeat the adjustment if necessary after 5 minutes until the reading on the display panel is stable. Turn on the injection switch (the injection light is on), adjust the set value to the required concentration, and make the concentration reach the set value concentration (the injection light is off), and maintain it for at least 10 minutes. After that, the content in the box is automatically adjusted by the controller.
(7) To adjust the humidity of the CO2 incubator, the method is to press the humidity adjustment knob first, then turn the humidity adjustment knob to the required cultivation humidity, and then automatically adjust the humidity by the humidity regulator.
2. Precautions for using CO2 incubator
(1) The incubator should be managed by a special person. Once any switches and adjustment knobs on the operation panel are fixed, do not twist them at will, so as not to affect the fluctuation of temperature, CO2 and humidity in the incubator, and reduce the sensitivity of the machine.
(2) The water added needs to be distilled water or deionized water to prevent the minerals in the water tank from depositing and corroding. The water is changed once a year. Check frequently that there is enough water in the tank.
(3) The cabinet should be scrubbed and disinfected with disinfectant regularly, and the shelf can be taken out for cleaning and disinfection to prevent other microorganisms from contaminating and causing the experiment to fail.
